چکیده

Cisplatin (DDP)‑based chemotherapy is a standard treatment for cervical cancer, although resistance remains major concern. Hypoxia‑inducible factor‑2 α (HIF‑2α) plays an important role in resistance. MicroRNAs (miRs) can inhibit gene expression by binding to the 3'‑untranslated region of target gene. The authors' previous study showed that miR‑519d‑3p regulation HIF‑2α under hypoxic conditions cancer. However, function and regulatory mechanisms miR‑519d‑3p/HIF‑2α axis DDP‑resistance cancer are not fully understood. Therefore, aim present was investigate whether increased DDP regulating PI3K/AKT signaling pathway. It found lower DDP‑resistant cells (CaSki/DDP HeLa/DDP) compared with CaSki HeLa conditions. Additionally, overexpression decreased IC50 value CaSki/DDP HeLa/DDP cells, inhibited protein pathway Furthermore, it demonstrated reduced effect on cells. Moreover, results suggested also indicated suppressing

Knockdown of HMGN5 increases the chemosensitivity of human urothelial bladder cancer cells to cisplatin by targeting PI3K/Akt signaling

High-mobility group nucleosome-binding domain 5 (HMGN5) is the latest member of the HMGN family of proteins. Numerous studies have confirmed the carcinogenic role of HMGN5 in cancer, but its function in the regulation of chemosensitivity is largely unknown and controversial. A previous study by the authors of the present study demonstrated that HMGN5 contributes to the progression of urothelial...

Silencing RIF1 decreases cell growth, migration and increases cisplatin sensitivity of human cervical cancer cells

Replication timing regulatory factor 1 (RIF1) plays an important role in DNA replication regulation, stem cell pluripotency and DNA repair pathway. However, little is known about the molecular mechanisms and physiological significance of RIF1 in cancer and chemotherapy efficacy. In this study, we found that RIF1 is upregulated in cervical cancer tissues compared with normal tissues both at mRNA...

Short hairpin RNA targeting Twist1 suppresses cell proliferation and improves chemosensitivity to cisplatin in HeLa human cervical cancer cells

Development of multidrug resistance (MDR) remains a major hurdle to successful cancer chemotherapy and MDR1/P-gp overexpression is believed to be mainly responsible for MDR of tumor cells. Twist1, which is a highly conserved transcription factor that belongs to the family of basic helix-loop-helix proteins, has been shown to be a major regulator of the epithelial-mesenchymal transition (EMT), a...

Knockdown of CABYR-a/b increases chemosensitivity of human non-small cell lung cancer cells through inactivation of Akt.

UNLABELLED CABYR is a calcium-binding tyrosine phosphorylation-regulated protein that was identified as a novel cancer testis antigen in lung cancer in our previous study. However, the role of CABYR as a driver of disease progression or as a chemosensitizer is poorly understood. This study sought to investigate the relationship between the expression levels of CABYR-a/b, which are the two predo...
